Effect of Ethanolic Crude Extracts of Kam Lang Seu Krong (Strychnos axillaris) and Other 4 Thai Herbs on CYP3A4 Enzyme Activity

Abstract

   

This research was aimed to examine the effect of ethanolic crude extracts of 5 Thai herbs, Kam Lang Seu Krong (Strychnos axillaris), Pak Chiang Da (Gymnema inodorum (Lour.) Decne.), Ngeok Pla More (Acanthus ebracteatus), Pet Sang Kart (Cissus quadrangulars) and Jet Ta Mool Pleang Dang (Plumbago indica) on CYP3A4 enzyme function. Metabolic tests were carried out in vitro using rat liver microsome. Testosterone (TST) was used as a specific CYP3A4 probe substrate.  The amount of major metabolite of TST was detected by using High Performance Liquid Chromatography (HPLC).  It was found that the ethanolic crude extract of Pak Chiang Da, Ngeok Pla More, Pet Sang Kart, Kam Lang Seu Krong and Jet Ta Mool Pleang Dang inhibited CYP3A4 enzyme activity in wide range (29.11, 21.8, 17.63, 62.79 and 61.09 % inhibition respectively).  The same tests with hexane and aqueous extracts of these herbs are in progress.
